Lineage 13 regurgitate samples were collected from brown noddies (Anous stolidus) in December 2017 and January 2019 on Egg Island, St Helena by St Helena Government (SHG). Details of samples are kept at EMD. 4 brown noddy samples (squid beaks) were sent to project parnters in France and Portugal for processing and identification. The other 9 noddy samples were sent to project partners at Cardiff University and used for DNA barcoding analysis to identify prey items. Primer pair LC01490_CephF & HC02198_CephR were used to amplify the upstream region of the mitochondrial COI gene. The downstream region of 16S rRNA was amplified using primer pair 16Sar and 16Sb. Sequences were then amplified and identified to species level through BLAST.
